Descrição da vaga: Offer DescriptionDysfunction in transcription factor (TF)-mediated signaling is associated with metabolic diseases. Searching for TFs and their regulatory sequences has supported studies of gene regulation in different physiological processes. We first analyzed large-scale datasets. The analysis revealed reduced expression of the TF Nr2f6 in skeletal muscle of cold-exposed mice. Then, Nr2f6 was silenced in C2C12 cells and RNAseq analysis was performed. The terms associated with the processes Metabolism and Ca2+ transport were upregulated, highlighting the role of Nr2f6 in metabolism. Skeletal muscle increases heat production by uncoupling ATP hydrolysis with Ca2+ transport by the Serca protein, indicating a role of Nr2f6 in muscle thermogenesis. Genes associated with thermogenesis including the ATP/ADP transporter and ryanodine receptor were also upregulated in Nr2f6-silenced cells. We then found Nr2f6-responsive elements in regulatory sequences of Ca2+ transport genes. These findings suggest that the nuclear receptor, Nr2f6, may be an important regulator of mitochondrial function and thermogenic processes in skeletal muscle.This opportunity is open to candidates of any nationality.
